چکیده

The rate of mitoehondrial carnitine-carnitine exchange mediated by carnitine acylcarnitine translocase was measured by föllowing the uptake of L-[methyl-C]carnitine. It was demonstrated that the hypoglycaemic compound 2-(3^methyKcinnamyl-hydrazono)-propionate causes a concentration-dependent decrease in the rate of the translocase-mediated transport of carnitine in guinea pig liver mitochondria. Apparent initial infhix rates were decreased by 20% at 0.3 mmol/1 2-(3-methyl-cinnamyl-hydrazono)-propionate, 38% at 0.5 mmol/1, and 75% at 2.0 mmol/1 of this compound. This finding may explain the previously observed inhibitory effects öf this substance on long^chain fatty acid oxidation, ketone body production and gluconeogenesis.

Characterization of carnitine acylcarnitine translocase system of heart mitochondria.

Mersalyl inhibited the respiration of heart mitochondria under conditions that required the transport of (-)-carnitine and acyl(-)-carnitines. The exchange of external carnitine and acylcarnitines for intramitochondrial carnitine was also inhibited by mersalyl and 1 mM mersalyl proved suitable for the inhibitor-stop assay of carnitine acylcarnitine translocase. The carnitine-carnitine and (-)-c...

Phytanic acid and pristanic acid are oxidized by sequential peroxisomal and mitochondrial reactions in cultured fibroblasts.

The relationship between peroxisomal and mitochondrial oxidation of the methyl branched fatty acids, phytanic acid and pristanic acid, was studied in normal and mutant human skin fibroblasts with established enzyme deficiencies. Tandem mass spectrometry was used for analysis of the acylcarnitine intermediates. In normal cells, 4,8-dimethylnonanoylcarnitine (C11:0) and 2,6-dimethylheptanoylcarni...

A mitochondrial carnitine acylcarnitine translocase system.

Acetylation of added (-)carnitine by heart mitochondira coupled to the oxidation of pyruvate in the presence of malonate was inhibited, apparently competitively, by long chain acyl(+)carnitines although the activity of carnitine acetyltransferase (EC 2.3.1.7) itself was not affected. Mitochondria have been found to possess a translocase system that allows the transport of carnitine and acylcarn...

Pharmacokinetic analysis of the cardioprotective effect of 3-(2,2, 2-trimethylhydrazinium) propionate in mice: inhibition of carnitine transport in kidney.

The site of action of 3-(2,2,2-trimethylhydrazinium) propionate (THP), a new cardioprotective agent, was investigated in mice and rats. I.p. administration of THP decreased the concentrations of free carnitine and long-chain acylcarnitine in heart tissue. In isolated myocytes, THP inhibited free carnitine transport with a Ki of 1340 microM, which is considerably higher than the observed serum c...

INHIBITION BY a-CYANO-4-HYDROXYCINNAMATE AND EVIDENCE FOR SEPARATE IDENTITY FROM THE PYRUVATE TRANSPORTING SYSTEM OF MITOCHONDRIA*

Some of the known inhibitors of pyruvate transport inhibited the activity of carnitine-acylcarnitine translocase. Their order of effectiveness, with millimolar concentration required for 50% inhibition given in parentheses, was: Compound UK-5099 (cz-cyano+(l-phenylindol-3-yljacrylate) (0.1); cu-cyano-4-hydroxycinnamate (0.17); a-cyano-3-hydroxycinnamate (1); cu-cyanocinnamate (1); cu-fluorocinn...
